Activity of glucose-fructose oxidoreductase in fresh and permeabilised cells of Zymomonas mobilis grown in different glucose concentrations BJM
Erzinger,Gilmar Sidnei; Silveira,Mauricio Moura da; Costa,José Paulo Castilho Lopes da; Vitolo,Michele; Jonas,Rainer.
Previously grown cells of the ethanologenic bacterium Zymomonas mobilis produce sorbitol and gluconic acid, in reactions catalysed by the periplasmic enzymes glucose-fructose oxidoreductase (GFOR) and glucono-delta-lactonase (GL). The GFOR/GL system activity, in cells to be used in this bioconversion, depends on growth conditions. In batch runs, with initial glucose concentrations (S0) from 42 to 230 g/L, the highest specific and total GFOR/GL activities were obtained with S0 = 153 g/L (12.6 U/g cells and 62 U/L). Higher S0 led to decreasing activities in fresh cells. With S0 = 209 g/L, the final specific activity was only 7.0 U/g. After disruption of cells, however, an activity over 15 U/g was revealed. Since growth inhibition with S0 over 153 g/L was...

ABSTRACT This study investigated the anesthetic potential of the essential oil (EO) of Aloysia polystachya in juveniles of dusky grouper (Epinephelus marginatus). Fish were exposed to different concentrations of EO of A. polystachya to evaluate time of induction and recovery from anesthesia. In the second experiment, fish were divided into four groups: control, ethanol and 50 or 300 µL L−1 EO of A. polystachya, and each group was submitted to induction for 3.5 min and recovery for 5 or 10 min. The blood gases and glucose levels showed alterations as a function of the recovery times, but Na+ and K+ levels did not show any alteration. In conclusion, the EO from leaves of A. polystachya is an effective anesthetic for dusky grouper, because anesthesia was...

Silva,Lenise de Lima; Garlet,Quelen Iane; Koakoski,Gessi; Abreu,Murilo Sander de; Mallmann,Carlos Augusto; Baldisserotto,Bernardo; Barcellos,Leonardo José Gil; Heinzmann,Berta Maria.
ABSTRACT The aim of this study was to evaluate the anesthetic activity of the essential oil (EO) of Ocimum americanum L. in silver catfish (Rhamdia quelen ). In the first experiment, the depressor effects and chemical composition of the leaf EO (LEO) and inflorescence EO (IEO) were compared. Juveniles (n = 10) were placed in aquaria containing different concentrations of EO (25 - 500 mg L-1) to determine the point at which anesthesia was induced and the length of the recovery period. In the following experiment, the effects of 300 and 500 mg L-1 LEO exposure on stress parameters (plasma cortisol, glucose and sodium levels) after air exposure for 1 min were assayed. Fish (n = 10 per sampling time) were sampled immediately or transferred to anesthetic-free...

Approches cellulaires et moléculaires de la gestion des réserves chez l’huître, Crassostrea gigas (Thunberg, 1793). Suivi saisonnier du métabolisme énergétique et effet du niveau trophique sur la mise en réserve ArchiMer
Hanquet, Anne-caroline.
In the Pacific oyster, Crassostrea gigas, the energy necessary for general metabolism and gametogenesis is mainly provided by the glycogen stored in the vesicular cells. The first level of glycogen metabolism control is the entrance of glucose into cells. The monitoring of the uptake capacity of [U-14C] D-glucose revealed the existence of a saturable component with kinetic constants: Km = 0.71 mM ; Vmax = 0.22 nmol/h/106cells. The use of inhibitors has revealed the presence of carriers belonged to GLuT and SGLT types. A follow-up showed a seasonal variation of the uptake capacity and incorporation of glucose into glycogen, and a high expression of SGLT during mobilization of glycogen and gametes maturation. A bioassay in a controlled rearing system has...

Association between heart rate, heart rate variability, cortisol, glucose and electrolytes in healthy newborn calves Arq. Bras. Med. Vet. Zootec.
Quevedo,D.A.C.; Lourenço,M.L.G.; Bolaños,C.A.D.; Takahira,R.K.; Oba,E.; Alfonso,A.; Chiacchio,S.B..
ABSTRACT This study aims at evaluating the effects of electrolytes, glucose and cortisol levels over heart rate (HR) and heart rate variability (HRV) in healthy newborn calves. Seventeen healthy Holstein calves were evaluated during their first month of life, and the plasma concentrations of glucose, cortisol, calcium, magnesium, inorganic phosphorus, sodium and potassium were analyzed. HRV indexes were determined in the time and frequency domains through the analysis of neonatal electrocardiogram recordings. In the first day, low blood levels of phosphorus presented a strong correlation with the HR and the increased high-frequency components of HRV. The plasma concentrations of magnesium decreased significantly throughout the 35 days, revealing a positive...

Azadirachtin biosynthesis induction in Azadirachta indica A. Juss cotyledonary calli with elicitor agents BABT
Rodrigues,Marcelo; Festucci-Buselli,Reginaldo Alves; Silva,Luzimar Campos; Otoni,Wagner Campos.
The use of cell and plant tissues culture techniques to produce economically important active metabolites has been growing. Among these substances, azadirachtin (AZA), produced by the neem tree (Azadirachta indica), has received considerable attention due to its bioinsecticide action. The main goal of this work was to analyze the AZA levels in neem cotyledonary calli. The calli were grown in agitated Woody Plant Medium (WPM) liquid medium, supplemented with glucose (Gl), hydrolyzed casein (HC) and methyl jasmonate (MeJ) as elicitor agent. An interaction was observed between these substances, depending on in vitro cultivation time with orbital agitation. The highest concentrations (average of 0.2470 µg g-1) of AZA were produced in the first and second weeks...

Biochemical and morphological changes during the growth kinetics of Araucaria angustifolia suspension cultures BABT
Santos,André Luis Wendt dos; Silveira,Vanildo; Steiner,Neusa; Maraschin,Marcelo; Guerra,Miguel Pedro.
Embryogenic cultures of Araucaria angustifolia were established in a BM liquid medium supplemented with 2 µM 2,4-dichlorophenoxyacetic acid, 1 µM 6-benzylaminopurine and 1 µM kinetin (BM2) and in a BM medium free of growth regulators (BM0). During 42 days in culture, the cell growth pattern of both cultures was similar. The pH of the culture medium of both BM0 and BM2 underwent progressive reduction during culture time. For both the embryogenic cultures a preferential uptake of glucose in the late stages of cell growth kinetics was observed. The extracellular protein content was similar for both the embryogenic cultures. Acetocarmine and Evan's blue double stain showed major differences for early somatic embryo organisation, in which only the embryogenic...

Biochemical profile of amniotic fluid for the assessment of fetal and renal development BJMBR
Oliveira,F.R.; Barros,E.G.; Magalhães,J.A..
Creatinine plays a key role in the function and maturation of fetal kidneys throughout pregnancy. It is important to identify other markers that may help in the diagnosis of renal dysfunction. Our aim was to determine the profile of and the correlation between biochemical markers to be used to assess renal function and maturation of the fetus in the amniotic fluid during pregnancy and to determine the distribution of normal values for creatinine, N-acetyl-ß-D-glucosaminidase (NAG), ß2-microglobulin, glucose, urea, sodium, potassium, phosphorus, calcium, uric acid, albumin, and osmolality in three gestational age groups. This was a cross-section study that assessed 115 samples of amniotic fluid during three different periods of pregnancy, i.e., 13 to 20, 27...

Blood metabolites as predictors to evaluate the body condition of Neopelma pallescens (Passeriformes: Pipridae) in northeastern Brazil Rev. Bras. Zool.
Azeredo,Luane M.M.; Oliveira,Talita C.; Lopez,Luiz C.S..
ABSTRACT Body condition is an important ecological attribute that can provide a measure of the energy stored by an animal or population. Body condition is traditionally ascertained with morphometric body condition indices (BCIs), but some researchers have used various physiological and biochemical parameters as predictors of body condition. This study aimed to test reliable biochemical predictors of body condition in individuals of Neopelma pallescens (Lafresnaye, 1853), found in an Atlantic forest patch. For this purpose we examined the relationship between body condition and the concentrations of blood glucose and ketones. We also verified correlations between blood metabolite concentrations and ecological variables, including time of day, seasons,...

Carbohydrate metabolism and metabolic disorders in horses R. Bras. Zootec.
Hoffman,Rhonda M..
Horses evolved consuming primarily fermentable forage carbohydrates, but forage diets have been traditionally supplemented with grain meals rich in starch and sugar in order to provide additional calories, protein and micronutrients. Starch and sugar are important for performance horses, but the consumption starch-rich meals may cause equine digestive and metabolic disorders. The critical capacity for preileal starch digestibility appears to be 0.35 to 0.4% but may be as little, depending on the source of starch. Small intestinal absorption of simple sugars is limited by the activity and expression of two classes of glucose carrier proteins, which are affected by chronic intake of hydrolyzable carbohydrate but may be sluggish to respond to abrupt changes...

Classification of Bovine Follicles Based on the Concentrations of Steroids, Glucose and Lactate in Follicular Fluid and the Status of Accompanying Follicles OAK
NISHIMOTO, Hiromi; HAMANO, Seizo; HILL, Glen Alan; MIYAMOTO, Akio; TETSUKA, Masafumi; 宮本, 明夫; 手塚, 雅文.
A simple and clear means to identify the physiological status of follicles is essential for study of follicular biology. In the present study, we verified a novel classification procedure based on analysis of the follicular population and glucose concentration in follicular fluid (FF) as an alternative method to classify bovine follicles. Paired ovaries were collected from heifers, and the number of follicles and stage of the CL were recorded. Follicles were initially divided into the following 3 groups according to diameter and the ratio of E2 and P4 (E/P): E2 active (E-A: E/P≥1), E2 inactive (E-I: E/P<1, ≥8.5 mm) and small follicles (E/P<1, <8.5 mm). E-A follicles were easily identified as E2-rich dominant follicles and were further classified...

Determination of optimal glucose concentration for microcalorimetric metabolic evaluation of equine spermatozoa BABT
Vasconcelos,André Belico de; Souza,Patrícia Castanheira de; Varago,Fabiana Cristina; Lagares,Monique de Albuquerque; Santoro,Marcelo Matos.
The heat conduction microcalorimeter can be used to evaluate the metabolic rates of the sperm cell. Two ejaculates of four stallions were cooled to +5ºC and checked for sperm motility (bright field microscopy), viability (eosin 3%), functional membrane integrity (hyposmotic swelling test), and heat production (microcalorimetry). Glucose and sperm cell concentrations were determined in order to measure the heat outputs resulting from sperm metabolism. Sperm viability, membrane integrity and sperm motility did not differ among the different glucose concentrations tested. Nevertheless, the highest heat output detected by the microcalorimeter was obtained with 6 mM glucose and 10(8) spermatozoa/mL. Since conduction microcalorimetry offered additional...

Development of the insulin secretion mechanism in fetal and neonatal rat pancreatic B-cells: response to glucose, K+, theophylline, and carbamylcholine BJMBR
Mendonça,A.C.; Carneiro,E.M.; Bosqueiro,J.R.; Crepaldi-Alves,S.C.; Boschero,A.C..
We studied the development of the insulin secretion mechanism in the pancreas of fetal (19- and 21-day-old), neonatal (3-day-old), and adult (90-day-old) rats in response to stimulation with 8.3 or 16.7 mM glucose, 30 mM K+, 5 mM theophylline (Theo) and 200 µM carbamylcholine (Cch). No effect of glucose or high K+ was observed on the pancreas from 19-day-old fetuses, whereas Theo and Cch significantly increased insulin secretion at this age (82 and 127% above basal levels, respectively). High K+ also failed to alter the insulin secretion in the pancreas from 21-day-old fetuses, whereas 8.3 mM and 16.7 mM glucose significantly stimulated insulin release by 41 and 54% above basal levels, respectively. Similar results were obtained with Theo and Cch. A more...
